GIII Series™ Family Summary
and Features

 

To facilitate homeostasis for gametes and embryos the GIII Series™ has been developed. The media of the GIII Series™ all have the same ionic backbone resulting in similar osmolality from oocyte aspiration and sperm preparation to embryo culture and transfer.

 

All IVF media of the GIII Series™, with the exception of G-RINSE™ and EmbryoGlue®, require albumin supplementation either with G-MM™ or HSA-solution™.

As a blood product, HSA is a risk factor in embryo culture. Even though albumin is tested for HIV, hepatitis A, B and C, there is no reliable test for prion transmitted disease such as Creutzfeld Jacobs disease. To circumvent the risks of HSA, recombinant human albumin (r-HA) has been recently introduced for media supplementation.

G-MM™ contains recombinant human albumin in the concentration of
50 mg/mL. HSA-solution™ is human serum albumin in the concentration of 100 mg/mL.

Oocyte retrieval and handling media
Before oocyte retrieval, the cervix and utensils should be washed using G-RINSE™.

For oocyte retrieval and for handling of gametes and embryos G-MOPS™ has been designed. G-MOPS™ contains the carbohydrates and amino acids that are required to support the gametes and embryos and contains a buffer to maintain pH in an air atmosphere. After oocyte identification, the oocytes should be rinsed in G-MOPS™ and transferred to G-FERT™.

Sperm preparation media

For sperm preparation, G-SPERM™ should be used. G-SPERM™ is designed to support the washing and purification of sperm. G-SPERM™ contains Gentamicin, the carbohydrates required to support sperm function and Hyaluronan. After washing, the sperm should be diluted in G-FERT™ for fertilization.

Fertilization Media

G-FERT™ is designed to support the preparation of gametes and in vitro fertilization. G-FERT™ contains the carbohydrates and amino acids to support both male and female gametes. G-FERT™ also contains Hyaluronan.

Optimized culture conditions

After fertilization, continued culture takes place in G-1™ version 3 until transfer on day 2 or 3. G-1™ version 3 is designed to support the development of cleavage stage embryos to around the 8-cell stage. G-1™ version 3 contains Hyaluronan, carbohydrates, amino acids and chelators to support the early embryo.

For continued culture to the blastocyst stage G-2™ version 3 should be used. G-2™ version 3 is designed to support the development of the embryo from around the 8-cell stage to the blastocyst stage. G-2™ version 3 contains Hyaluronan, carbohydrates, amino acids and vitamins to support the later stage embryo.

For optimal embryo culture, droplets under oil are preferred. For covering of droplets G-OIL™ is recommended. G-OIL™ is liquid paraffin oil designed for use as an overlay for media.

New embryo transfer medium

For embryo transfer EmbryoGlue® has been designed. EmbryoGlue® contains carbohydrates, amino acids, vitamins and Hyaluronan.

G-RINSE™ IVF Media

Solution for rinsing of contact materials and for washing of the cervix. Not for culture.

Characteristics
No protein supplementation needed.

G-MOPS™

Medium for handling and manipulating oocytes and embryos in ambient atmosphere.

Characteristics
Contains NEAA and G, P, L and MOPS.

No pre-equilibration in CO2 needed. No protein supplementation needed for oocyte retrieval. For embryo manipulation after the addition of G-MM™ or HSA-solution™.

G-SPERM™

Medium for sperm preparation.

Characteristics
Contains HA, NEAA, G, P, L and Gentamicin

For use after the addition of G-MM™ or HSA-solution™.

G-FERT™

Medium for preparation and handling of gametes and for
in vitro fertilisation.

Characteristics
Contains HA, NEAA, G, P and L.

For use after the addition of G-MM™ or HSA-solution™.

G-1™ version 3 Media

Medium for culture of embryos from the pronucleate stage to day 2 or day 3.

Characteristics
Contains HA, EDTA, NEAA, G, P and L.

Lower G, higher P and L to meet demand of pre-compaction stage, NEAA facilitate embryo development and viability, Hyaluronan to improve embryo cryosurvivability.

For use after the addition of G-MM™ or HSA-solution™.

G-2™ version 3

Medium for culture of embryos from day 3 to the blastocyst stage.

Characteristics
Contains HA, NEAA, EAA, Vitamins, G, P and L; EDTA not included.

Higher G, lower P and L to meet demand of post-compaction stage, NEAA, EAA, Vitamins facilitate blastocyst development, differentiation, metabolism and viability; EDTA omitted to improve blastocyst formation, Hyaluronan to improve embryo cryosurvivability.

For use after the addition of G-MM™ or HSA-solution™.


G-PGD™

Medium for embryo biopsy. Not for culture.

Characteristics
Calcium and Magnesium free MOPS buffered medium to facilitate the embryo biopsy procedure.

Contains carbohydrates and NEAA to support the cleavage stage embryo.
No CO2 equilibration needed.

For use after the addition of G-MM™ or HSA-solution™.

HSA-solution™

Human Serum Albumin supplement.

Characteristics
Strictly controlled pharmaceutical Human Serum Albumin.

G-MM™

Recombinant Human Albumin supplement.

Characteristics
Ultra-high purity and LOT-to-LOT consistency enabling consistent good success rate.

G-FreezeKit Blast™

3 Media for freezing of blastocyst stage embryos.

Characteristics
MOPS buffered media containing the cryo-protectants glycerol and sucrose. No CO2 equilibration needed.

For use in sequence after the addition of G-MM™ or HSA-solution™.


G-ThawKit Blast™

4 Media for thawing of blastocyst stage embryos.

Characteristics
MOPS buffered media containing the cryo-protectants glycerol and sucrose. No CO2 equilibration needed.

For use in sequence after the addition of G-MM™ or HSA-solution™.
